Description / Action: 

CLINTABS Tablets contain clindamycin hydrochloride a semisynthetic lincomycin antimicrobial produced by a 7(S)-chlorosubstitution of the 7(R)-hydroxyl group of a naturally produced antibiotic produced by Streptomyces lincolnensis var. lincolnensis.

It is indicated for the treatment of infections caused by susceptible strains of the designated microorganisms and inhibits protein synthesis in the bacterial cell by binding with 50S sub-unit of the ribosome. It binds to the soluble RNA fraction of certain ribosomes which results into inhibition of the binding of amino acids to those ribosomes and causes irreversible modification of the protein-synthesizing subcellular elements at the ribosomal level.

Clindamycin has activity against a wide variety of aerobic and anaerobic bacterial pathogens and a bacteriostatic action.

Clindamycin is rapidly absorbed from the canine gastrointestinal tract, reaches at peak serum concentrations 1 hour and 15 minutes after oral administration and excreted in urine and feces (elimination half-life is approximately 5 hours in healthy dogs).

Indication / Uses:

CLINTABS Tablets are indicated in the conditions and infections caused by micro-organisms listed below:
• Skin Infections (wounds and abscesses) caused by coagulase positive staphylococci (Staphylococcus intermedius or Staphylococcus aureus).
• Deep wounds and abscesses caused by Bacteroides fragilis, Fusobacterium necrophorum, Prevotella melaninogenicus, and Clostridium perfringens.
• Dental infections caused by Staphylococcus aureus, Bacteroides fragilis, Prevotella melaninogenicus, Fusobacterium necrophorum and Clostridium perfringens.
• Osteomyelitis caused by Bacteroides fragilis, Staphylococcus aureus, Fusobacterium necrophorum, Prevotella melaninogenicus and Clostridium perfringens.

Dosage and administration:

Infected Wounds, Abscesses, and Dental Infections

Oral: 2.5-15.0 mg/lb body weight q12h or twice daily.

Duration: maximum upto 28 days if clinical judgment indicates. Treatment should not be continued for more than 3 or 4 days in case of acute infections if no response to therapy is seen.

Dosage Schedule:

CLINTABS 25 mg, administer 1-6 tablets for each 10 pounds of body weight twice daily
CLINTABS 75 mg, administer 1-6 tablets for each 30 pounds of body weight twice daily
CLINTABS 150 mg, administer 1-6 tablets for each 60 pounds of body  weight twice daily

Osteomyelitis:

Oral: 5.0-15.0 mg/lb body weight twice daily.

Duration: for a minimum of 28 days. If the response to therapy is not seen then treatment should not be continued for longer than 28 days.

Dosage Schedule:

CLINTABS 25 mg, administer 2-6 tablets for each 10 pounds of body weight twice daily.
CLINTABS 75 mg, administer 2-6 tablets for each 30 pounds of body weight twice daily.
CLINTABS 150 mg, administer 2-6 tablets for each 60 pounds of body weight twice daily.

Species: Approved for Oral Use in Dogs Only.

Over-dosage / Side effects / Contraindications /Warnings:

Occasionally observed side effects in either clinical trials or during clinical use were vomiting and diarrhea in dogs.

Do not administer in pets with a known history of hypersensitivity to clindamycin or lincomycin.

Because of potential adverse GI effects, do not administer to guinea pigs, rabbits, horses, hamsters, chinchillas or ruminating animals.

Use with caution in where severe kidney or liver disease is present and in animals receiving neuromuscular blocking agents such as succinylcholine because clindamycin has neuromuscular blocking properties.

Patients with very severe renal disease and/or very severe hepatic disease accompanied by severe metabolic aberrations should be dosed with caution, and serum clindamycin levels monitored during high-dose therapy. Periodic liver and kidney function tests and blood counts should be performed if prolonged therapy of one month or greater is going on,

Administration clindamycin should be avoided in those species sensitive to the gastrointestinal effects of clindamycin because it occasionally results in overgrowth of non-susceptible micro-organisms such as yeast and clostridia. Appropriate measures should be taken In case of reported superinfections, as indicated by the clinical situation.

Discontinue the treatment of acute infections if there is no response to the therapy for more than three or four days. Haematobiochemical analysis should be done /monitored in animals on either high dose or prolonged therapy.

Keep out of reach of children. Not for human use. Keep this reaching out of the children.

Safety: Age / Pregnancy/ Withdrawal:

Safety in breeding males or pregnant females has not been established.

Rat and Dog Data: rats and dogs during one year oral toxicity studies  at doses of 30, 100 and 300 mg/kg/day (13.6, 45.5  and 136.4 mg/lb/day) have shown to be well tolerated. There was no differences in the parameters evaluated to assess toxicity in comparison to contemporary controls.  Rats with dose rate of 600 mg/kg/day (272.7 mg/lb/day) for six months tolerated the drug well; however, vomiting, anorexia and subsequent weight loss was seen in dogs at dose rate of 600 mg/kg/day (272.7 mg/lb/day) on oral administration. These dogs had erosive gastritis and focal areas of necrosis of the mucosa of the gall bladder.
