Tacoline Ointment contains tacrolimus, a macrolide immunosuppressant produced by Streptomyces tsukubaensis. It is for ophthalmic use only.
Clinical Pharmacology:
Tacrolimus (formerly FK-506) is an immunostimulatory drug with a mechanism that is comparable to cyclosporine A but substantially more potent (up to 100 times). Its action is initiated by binding to a class of peptidyl-prolyl cis-trans isomerases (PPlases), designated FK506- binding proteins (FKBPS). The FKBP-12 protein, which has a cytosolic mass of about 12 kDa, is the main FKBP found in T lymphocytes. Moreover, it has been claimed that tacrolimus blocks mast cell histamine release and reduces prostaglandin synthesis in a de novo manner.
Tacrolimus has anti-inflammatory properties and reduces the release of pro-inflammatory cytokines, activated T lymphocytes, and markers of apoptosis. It also has neuroprotective properties and prevents the loss of conjunctival epithelium and the reduction in goblet cell numbers. Many chronic ocular disorders share similar mechanisms, and the effects of tacrolimus on corneal grafts, inflammatory conjunctival and corneal diseases like Keratoconjunctivitis Sicca.
In order to treat these corneal and ocular surface diseases, ophthalmic tacrolimus is a welcome addition to the therapeutic toolbox.

Indication / Uses:
Used in treatment and management of Keratoconjunctivitis Sicca.
Dosage and administration:
Apply ¼ inch strip of ointment. Apply twice daily or as directed by Veterinarians.
Directions for use:
Unscrew the white cap and discard the spacing ring. Refit the white cap firmly down to puncture the diaphragm. Remove the small-cap and squeeze the tube for use.
Do not use any other outside article to puncture the diaphragm. Replace the cap tightly after use.
Do not touch the tip or other dispensing tip to any surface since this may contaminate the ointment.
Sterile until opened. Use within one month after opening the tube.
Overdoses / Side effects / Contraindications /Warnings:
TACOLINE Ointment is contraindicated in dogs with a history of hypersensitivity to Tacrolimus.
If irritation persists or increases discontinue the use and consult your veterinarian.

Pharmaceutical precautions / Instructions:
Store in a cool & dry place at a temperature not exceeding 25°C. Do not freeze.
