Description / Action:

FIROtail tablets contain firocoxib. Firocoxib is a non-steroidal anti-inflammatory drug for the management of pain and inflammation in dogs. It is a selective COX-2 inhibitor with lesser side effects. Firocoxib is also indicated in horses but this preparation is strictly for dogs.

Pharmacokinetics:

When given orally to adult dogs who have fasted, firocoxib has an approximate 38% absolute bioavailability at a dose of 5 mg/kg. Firocoxib is rapidly cleared from the blood via hepatic metabolism and fecal excretion. Despite a high level of plasma protein binding (96%), firocoxib exhibits a large volume of distribution and a terminal elimination half-life of 7.8 hours. Subjects’ oral medication absorption processes differ greatly from one another. Co-administration of firocoxib with food delays drug absorption (Tmax from 1 to 5 hours) and decreases peak concentrations (Cmax from 1.3 to 0.9 mcg/mL). But at the prescribed dosage, food has no effect on the total oral bioavailability.

Class: Non-steroidal anti-inflammatory drug (NSAID)

Indication / Uses:

Firotail (firocoxib) Chewable Tablets are indicated for

• Management of pain and inflammation associated with osteoarthritis and
• Management of postoperative pain and inflammation associated with orthopaedic and soft-tissue surgeries in dogs.

Dosage and administration:

The recommended dose of firocoxib is 5 mg/kg/day or as directed by the veterinarian.

Carefully consider the potential benefits and risks of Firotail and other treatment options before deciding to use Firotail. Use the lowest effective dose for the shortest duration consistent with individual response. The recommended dosage of Firotail (firocoxib) for oral administration in dogs is 5.0 mg/kg body weight once daily as needed for osteoarthritis and for 3 days as needed for postoperative pain and inflammation associated with soft-tissue and orthopedic surgery. The dogs can be treated with Firotail approximately two hours prior to surgery. The tablets are scored and dosage should be calculated in half tablet Increments. Cyclooxygenase inhibitory NSAIDs as a class may be harmful to the kidneys, liver, and gastrointestinal tract or cause toxicity. Each patient has a different sensitivity to side effects related to drugs. Dogs those have shown adverse reactions from one NSAID may experience the same from another NSAID also. Individuals who are dehydrated, on diuretics concurrently, or who already have renal, cardiovascular, or hepatic impairment are most vulnerable to negative effects. When administering potentially nephrotoxic medications concurrently, caution and close observation are needed. NSAIDS may inhibit the prostaglandins that TOM maintain normal homeostatic function. Such anti prostaglandin effects may result in clinically significant disease in patients with underlying or pre existing disease that has not been previously diagnosed. Since NSAIDs possess the potential to produce gastrointestinal ulceration and/or gastrointestinal perforation, concomitant use of Firotail Chewable Tablets with other anti- inflammatory drugs, such as NSAIDs or corticosteroids, should be avoided. The concomitant use of protein bound drugs with firocoxib has not been studied in dogs. Commonly used protein-bound drugs include cardiac, anticonvulsant, and behavioural medications. The influence of concomitant drugs that may inhibit the metabolism of Firotail Chewable Tablets has not been evaluated. Drug compatibility should be monitored in patients requiring adjunctive therapy.

If additional pain medication is needed after the daily dose of Firotail, a non- NSAID class of analgesic may be necessary. When using anaesthetics and NSAIDS simultaneously, use caution as anaesthetic medications have the potential to impact renal perfusion. When using NSAIDs perioperatively, parenteral fluids should be taken into consideration to reduce the risk of renal complications.

Side effects / Adverse Reactions:

Firocoxib like other drugs in its class, is not free from adverse reactions. Owners should be advised of potential for adverse reactions and be informed of the clinical signs associated with drug tolerance. Adverse reactions may include vomiting, diarrhoea, decreased appetite, dark or tarry stools, increased water consumption, increased urination, pale gums due to anaemia, or behavioural changes. Serious adverse reactions associated with drugs can occur, and in this case owners should discontinue the Firotail therapy and contact the veterinarian immediately.
