Description:
Alizin injection contains aglepristone a synthetic steroid with anti-progesterone properties. It is used for the induction of abortion in pregnant female dogs for the management of mismating. Usual dose for induction of abortion is 10 mg/kg body weight for dogs.
Pharmacodynamic Properties:
Aglepristone is a synthetic steroid with an anti-pregestational action by competing with progesterone at its receptors.
In vitro, it shows affinity 3 times greater than of progesterone for the progestagen receptor prepared from female dog uterus and 3.8 times and 9 times greater respectively in female rabbits and female cats.
In vivo, in pregnant female rats, a dose of 10 mg/kg subcutaneously induced abortion in all treated animals.
Pharmacokinetic Characteristics:
In the non-pregnant female dogs, after two injections of 10 mg/kg/day 24 hours apart, the maximum concentration approximately 61 ng/ml is reached after 2.5 days. The average residence time is approximately 6 days, which includes the average absorption time from the injection site.
Following a single dose of 10 mg/kg/day in non-pregnant female dogs, 60% of the administered dose is excreted during the first 10 days and approximately 80% is excreted within 24 days, primarily via the feces (approximately 90%).

Indication / Uses:
Pregnant female dogs for the induction of abortion up to 45 days after mating.
Dosage and administration:
10 mg of algepristone per kg of body weight per day, or 0.33 ml Of solution per kg of body Weight per day, in two injections 24 hours apart.
Inject subcutaneously into the neck.
After each administration, it is advisable to massage the injection area lightly.
In confirmed pregnant female dogs, partial abortion can be found in about 5% of cases. A follow up clinical examination is always recommended to confirm uterine vacancy. This should be performed 10 days after administration of Alizin and at least 30 days after mating.

Side effects / Adverse reactions (frequency and seriousness):
The administration of Alizin is likely to induce a fleeting manifestation of pain during the injection. A local inflammatory reaction at the injection site, which is painless and always reversible, may also be observed. The severity of the reactions observed is related to the volume injected.
In female dogs treated late, after 20-30 days of gestation, abortion is accompanied by the physiological manifestations of parturition, foetal expulsion, mild anorexia and breast congestion.
After an abortion induced by Alizin, an earlier return to heat is frequently observed (the heat-to-heat interval is shortened by 1 to 3 months)
In rare cases in the 24 hours following the injection of aglepristone cardio-respiratory signs (shock, pulmonary oedema, bradycardia, dyspnoea,…) associated or not with systemic signs (Hypersalivation, vomiting, pallor..) or nervous signs (prostration, ataxia, dyspnea,…) have been observed.
Overdose (symptoms, emergency procedures, antidotes), if necessary:
A subcutaneous treatment with 30 mg of aglepristone par kg twice 24 hours apart, did not cause any side effects, except for local inflammatory reactions related to the injected.
Ulceration may occur at the Injection site. This reaction is related to the volume injected. A scar may persist for up to 3 months after administration.

i) Special precautions for use in animals
If the volume to be injected is greater than 5 ml carry out 2 injections in two different places on the skin of the neck in order to limit the appearance of ulceration and scarring.
ii) Special precautions for the person administering the veterinary medicinal product to animals
This product must be handled with care by pregnant women or by women likely to become pregnant.
